Un ligand d'affinit\u00e9, sp\u00e9cifique d'un site de liaison sur la mol\u00e9cule cible, est coupl\u00e9 \u00e0 une matrice de chromatographie inerte. Dans des conditions de liaison appropri\u00e9es, les mol\u00e9cules cibles sont li\u00e9es au ligand d'affinit\u00e9 en fonction de sa sp\u00e9cificit\u00e9. Les solut\u00e9s non li\u00e9s sont lav\u00e9s \u00e0 travers la colonne. Les mol\u00e9cules cibles adsorb\u00e9es sont ensuite d\u00e9sorb\u00e9es et \u00e9lu\u00e9es de la colonne. Une purification de plusieurs milliers de fois peut \u00eatre obtenue gr\u00e2ce \u00e0 la grande s\u00e9lectivit\u00e9 des interactions d'affinit\u00e9.<\/p><p>Les r\u00e9sines d'affinit\u00e9 sp\u00e9cifiques \u00e0 un groupe (par exemple Tosoh Bioscience) lient les mol\u00e9cules partageant des caract\u00e9ristiques structurelles sp\u00e9cifiques. Si une plus grande sp\u00e9cificit\u00e9 est requise, il est \u00e9galement possible d'utiliser des ligands ayant une sp\u00e9cificit\u00e9 pr\u00e9cise pour la mol\u00e9cule cible.
